PharmacoGenetic Neuroimaging approach to the understanding of human emotion

Recent research has linked a genetic variation in the serotonin transporter (5-HTTLPR) with emotional processing and mood disorders. However, the molecular mechanism that mediates the effect of 5-HTTLPR on neural responses to emotion remains unknown. I will present my recent research that combined genetic neuroimaging, pharmacological manipulation and psychological paradigms targeting emotion to address the role of serotonin transporter (5-HTT) in the 5-HTTLPR effects on human emotion. The findings first indicate significant genotype differences between short/short and long/long homozygotes of 5-HTTLPR in the emotional neural circuit, including amygdala, insula, thalamus, ACC, etc. Moreover, acute blockade of 5-HTT activity by citalopram (a highly selective serotonin reuptake inhibitor, SSRI) decreased the hyperactivity to negative emotions in long/long homozygotes and the citalopram effect was more salient in long/long than short/short homozygotes. Our findings provide the first pharmacogenetic neuroimaging evidence for a causal link between 5-HTT and 5-HTTLPR effects on human brain activity underlying emotional processing. The work illustrates a new, promising approach to the understanding of the neurobiological mechanism underlying human emotion and has important implications in clinical treatment of mood disorders.
